Themes Of The Crucbile In Arthur Miller's, ?The Crucible,? many themes are expressed throughout the play. Themes are the undertone of the story. A theme of a book usually sets the mood and describes what is happening during the time that the story is written. ?The Crucible? has many themes that show how everything was and how everyone acted in the year of 1692, in Salem, Massachusetts. Mass hysteria is the most obvious theme in the story. Mass hysteria is represented everywhere trouble was. One example is at the end of act one when the girls are screaming, crying, and starting to accuse people of being with the devil. When this happens, everyone gets scared and calls the marshal. The marshal begins to arrest people and brings them to court. The whole reason mass hysteria broke out is because of Abigail. One vengeful accusation from Abigail to her rival, Elizabeth Proctor, turns the whole village upside down. This confusion and madness is one of the main reasons so many lives were taken at the trials. This becomes a place where reasonable human beings can become released in an environment that allows little opportunity for relaxing. Another theme during the play is how much religion ruled these peoples lives. The court was the main ruling body of justice and was run by the church. The concept of justice in 1692 is shown when Arthur Miller dedicates the entire third act to the courtroom. Abigail pressures the girls to lie in court in order to accuse everyone that they didn't get along with of witchcraft. The separation of church and government didn't exist in 1692 in Salem, Massachusetts. Theocracy meant that Massachusetts was to be governed by God's laws. But this mixing up of the laws of God and the laws of government set up the chaos of the Salem witch trials. Greed and revenge was another major aspect that was shown in the story. Several characters find profit in this mass hysteria and try to change some events for their own needs and well-being. Thomas Putnam gains land by having his daughter Ruth accuse his neighbors of witchcraft. Also, Abigail gets revenge on the Proctors when her affair with John has been turned off. Superstition was the biggest cause of these trials. There were no real witches in Salem. Without the superstitious belief in witchcraft, this tragedy would have never happened. Arthur Miller clearly explains the how people react to things they do not understand. These were examples why The Salem Witch Trials happened. The theme of the story keeps the plot going. The significance of Garibaldis contribution Essay Giuseppe Garibaldi was born in Nice, and, although he was a born a French citizen, he regarded himself as Italian. Despite the pressure placed upon him to enter the Church, he joined the Merchant Navy where a chance meeting with Mazzini altered the entire course of his life. One of Mazzinis greatest disciples was Garibaldi, who was inspired with nationalist fervour and patriotic enthusiasm. He quickly converted to share Mazzinis aim of a united Italy, but only an Italy united as a republic. A revolutionary plot in 1833 in Piedmont was introduced to cause mutiny in the army but it failed and Garibaldi was sentenced to death. Execution could not be carried out as Garibaldi left the country prior to the trial. He moved to Brazil and then on to Uruguay where his military ability was first displayed. He fought, defending Uruguay in an Austrian take-over, however, news of the Italian revolutions of 1848 reached him and he decided to return to his homeland. He did this with some out of date weapons and some of his colleagues. Word of his military campaigns in South America had by now reached the Italian States and upon his return, recruits assembled to join Garibaldis legion. Garibaldis contribution in South America had a profound effect on events later on in the fight for unification, as it was due to his military involvement that he came back to Italy a prominent figure. It was only due to his reputation that he was able to recruit so many men. Although Garibaldi was originally drawn to the cause of unification as one of Mazzinis supporters, he saw the powerful Piedmontese state and monarchy as a more realistic way of achieving unification. It was at this point that Garibaldi and his mentor Mazzini parted ways. Mazzini was only interested in a unified Italy as a republic whereas Garibaldi wanted unification by any means possible. This is also where Garibaldi shows contrasts with Cavour who was above all else Piedmontese. February 1849 saw the establishment of the Roman Republic. The Republic itself was short-lived; only lasted four months. Garibaldi played a role in resisting the Catholic countries assisting the Pope however, the defenders  were greatly outnumbered by the powers helping the Pope and the fall of Rome was inevitable. After the fall of the Republic, Garibaldi made an appeal for new recruits to fight the war against the stranger. Although he offered no provisions or promises, almost 5000 men were recruited. Despite this, on their journey to Venetia, 3500 were lost. Garibaldi escaped to Genoa where he was arrested and released on the condition that he leave Italy at once. Upon his release, he travelled to North America where he stayed until 1854. When he returned, Garibaldis significance is clearly visible. A revolt which began in 1860 in Sicily came to the attention of Garibaldi and he set sail for Sicily almost immediately. His unexpected conquests in Sicily and Naples precipitated the unification of most of Italy. When he reached Sicily, Garibaldi was a welcome fighter. His weak army of 1200 now reaching 3000 took Palermo forcing the garrison to return to Naples making Sicily his. In September 1860 Naples fell leaving Garibaldi as Dictator over the Kingdom of Naples. Cavour feared that Garibaldi would not stop and would attempt to take Rome so he advised Victor Emmanuel to take Rome first. When the two forces met, Garibaldi proclaimed Victor Emmanuel the first King of Italy Plebiscites held in the newly acquired land showed immense support for annexation by Piedmont. In 1861, Garibaldis aim of a united Italy came into being; however, it did not include Rome or Venetia. By way of unification under control of Piedmont, the Piedmontese constitution and legal system were extended over the whole country. Though Rome remained to be occupied by France, continuing pressure from Italian liberals came for it to be freed and incorporated into the new Italian Kingdom as the historical capital. Apparently, with the implicit support of Victor Emmanuel, Garibaldi set off with 3000 volunteers to conquer Rome. When Garibaldi reached Palermo, he was greeted by calls of Rome or death and suddenly, Victor Emmanuel disowned the march. Garibaldi and his men were shot at by local troops in Calabria and only 500 remained. The whole operation was a disaster for Garibaldi and an embarrassment for the government. In 1864, the Italian government agreed to protect Rome form attack and to move the Italian capital from Turin to Florence and in return  the French withdrew from Rome. Garibaldi saw this as a betrayal of the cause for unification and decided to make an attempt to recapture Rome. Garibaldi and his men began their march to Rome but he was arrested. Garibaldi attempted to re-join his army and did succeed; however, his planned revolution with the Roman people did not happen. This was the end of Garibaldis contribution to Italian history. There is evidence to support both theories, but we still don't know for sure that we are not the only kind of life out there. There are different space bodies that are most important to have life in them; some of these places are Mars, Europa, Titan, Enceladus, and Ganymede and Calysto. One of the main reasons why scientists strongly believe that there is a good chance of life in Mars is because there is sufficient facts to imply that it had running water on its surface millions of years ago. There’s also evidence that ice and water existed on this planet. This evidence supports scientists who believe that life existed at some point on Mars. This data was discovered when the images taken from the Mars Reconnaissance Orbiter captured some characteristics of the planet's surface. Scientists believe it was created by the melting ice. Also in May 2009, images of an unmanned spaceship showed what it seemed to look like drops on the surface that appeared to be built during the time of the mission of the spacecraft on Mars. Another place that has scientists believes life is a moon of Jupiter, Europa. Some o f them believe that there is a giant, active salty ocean beneath its surface. This indicates that there is significant activity underneath that frozen surface. Another sign of life in Europe includes formations that looked like icebergs emerging from its surface, suggesting that oceans once covered the planet before it froze. The third space body where scientists think there is life is on one of Saturn's moons, called Enceladus. The Effect of Solvent Exposure and Temperature on Beet Cell Membrane Integrity Introduction: Cell membranes are vital to proper cellular function †¢ Cell membranes consist of a lipid bilayer made up of phospholipids †¢ Phospholipids are amphipathic molecules meaning they have a dual nature in that they show both hydrophilic (water-loving) and hydrophobic (water-fearing) properties †¢ The amphipathic nature of phospholipids causes them to spontaneously form bilayers in water based solutions o The inner and outer surfaces of the bilayers are hydrophilic and can interact with the aqueous environment, while the core of the bilayer is hydrophobic and able to exclude water and other polar molecules o Because of these properties, phospholipids bilayers are able to serve multiple functions including: ? act as barriers to protect the cell from the outside environment ? house proteins and carbohydrates necessary for cell to cell and extracellular communication ? rganize and prov ide surface areas for metabolic reactions †¢ While the phospholipids bilayer is a highly organized system with multiple functions, it is also highly dynamic and can be functionally described using the fluid mosaic model (FMM) o FMM characterizes the phospholipids bilayer of the cell membrane as highly dynamic and fluid meaning that the position (location) of any given phospholipids in the membrane is not fixed but fluid; thus, individual phospholipids can change positions are needed based on the current conditions. o Similarly proteins and carbohydrates are generally mobile in the membrane as well. †¢ This experiment was aimed at investigating the conditions that affect cell membrane integrity. o Because cell membranes are part of a living system they are sensitive to environmental conditions o Like most biological systems, cell membranes show a relatively narrow range of function with respect to pH, temperature, and exposure to organic solvents †¢ The experimental mo del employed was red beets Red beet cells contain large, membrane bound central vacuoles that house a pigment, betacyanin o When cell and vacuole membranes are in tact, betacyanin in contained within the cell o If the cell and vacuole membranes are disrupted, betacyanin can leak in the surrounding solution turning it pink o Higher amounts of membrane damage will lead to more leakage of betacyanin pigment from the cells. The more pigment present the higher the absorbance reading. o Therefore, betacyanin release, as determined by measuring absorbance, serves as a proxy for quantifying membrane damage. †¢ In this experiment, we tested the effect of changing two environmental conditions on cell membrane integrity.First, we tested the effect of temperature on cell membranes o To accomplish this we exposed beet cells to temperatures ranging from -5(C to 70(C o Because cell membranes maintain integrity under a narrow range of conditions, we hypothesize that exposure to higher temperat ures (50-70(C) and extreme low temperatures (-5(C) will cause severe damage to the cell membranes which will result in higher amounts of betacyanin being released from the cells. †¢ We also tested the effect of exposing beet cells to two different solvents o Methanol and acetone are organic solvents capable of dissolving hydrophobic species such as phospholipids o Acetone is generally considered a stronger organic solvent compared with methanol o Thus, we hypothesize that exposure to acetone will cause more damage to beet cell membranes compared with exposure to methanol.Additionally, we hypothesize that higher concentrations of acetone will cause more damage than lower concentrations. Similarly, higher concentrations of methanol will also cause more damage. Results (the data numbers are for example only and meant to reflect the general trend in the class; data from individual groups will vary): †¢ Experiment 1: The effect of temperature on cell membrane integrity o We tes ted the effect of exposing beet cells to various temperatures ranging from -5(C to 70(C. (Question) o To do this, uniform pieces of beet were placed in empty test tubes then incubated at a specific temperature. (How the question was addressed) ?Following this water was added to the test tube and the cells were incubated for 20 minutes. ? After incubation, the water was transferred to a clean tube and absorbance at 425 nm was determined. o Absorbace readings were similar at temperatures of 5? C, 25? C, 40? C, 55? C, and 70? C (A425 0. 25-0. 30) (Figure 1) (Experimental data) o Incubation at -5? C showed a dramatic increase in absorbance (A460 = 0. 95) (Figure 1) (Experimental data) o We conclude from this data that temperatures ranging from 5-70? C have only minor effects on cell membrane integrity. In contrast; exposure of cells to a temperature below freezing causes much more membrane damage. (Conclusion) Experiment 2: The effect of solvent on cell membrane integrity o We tested wh ether exposure to two different organic solvents, methanol and acetone, disrupted cell membrane integrity (Question) o Beet cells were incubated in 10%, 25%, and 50% methanol or 10%, 25%, and 50% acetone. Additionally, additionally cells were also incubated in saline, a solvent that mimics physiological conditions. (How the question was addressed) ? Following exposure to solvent, membrane damage was determined by measuring absorbance. o Incubation in 10% or 25% methanol yielded absorbance readings of 0. 2 and 0. 25 respectively; however, incubation in 50% methanol showed an increase in absorbance to 0. 5. Figure 2) (Experimental data) o Incubation in 10% or 25% acetone produced higher absorbance readings compared to the same concentrations of methanol (0. 4 and 0. 42 respectively). Similar to 50% methanol, 50% acetone yielded the highest absorbance reading of 0. 8 (Figure 2). (Experimental data) o Taken together, these data show that acetone causes more membrane damage than methanol and that concentrations of 50% solvent are more damaging than lower concentrations. (Conclusion) Overall Conclusion: †¢ We hypothesized that temperatures outside of the normal physiological range would disrupt membrane integrity causing betacyanin to leak out of the cells. Our data shows that our hypothesis was partly correct, low temperature, -5? C, inflicts the most damage to cell membranes while higher temperatures, ranging from 55-70? C induce little damage when compared with physiological temperatures. †¢ We also hypothesized that exposure to high concentration organic solvents would disrupt membrane integrity and that acetone, being a stronger solvent, would show more damage than methanol. o In this case our hypothesis was support. Acetone showed more damage as measured by betacyanin release than methanol. Additionally, the highest concentration of each solvent showed the most damage.
